Pulmonary drug delivery has developed to an appealing option for the treatment of several lung diseases, which entail, for example, the delivery of high lung doses or the formulation of biopharmaceuticals. New inhaler inventions and formulation strategies for dry powder inhalation emerged. Among them was an interesting concept to prepare the formulation by freeze-drying and disintegrate the lyophilisate into inhalable particles by an air impact at the time of inhalation. The evaluation and possible further development of this new concept is the subject of this thesis. The main focus is thereby on the improvement of the formulation towards the delivery of high lung doses.
