The molecular chaperone Hsp90 facilitates the activation and maturation of several hundred client proteins in the cell. In this thesis, the following aspects of Hsp90 were investigated: The functional analysis of its charged linker; the analysis of the C-terminal domain of the Hsp90 cochaperone p23 with respect to the independent functionality as well as its Hsp90-related functions; the effects of methylation as a new posttranslational modification of Hsp90 and folding of the client protein P1 by Hsp90.
