Engineering of riboswitch for metabolic pathway control and improved L–lysine production in Corynebacterium glutamicum

Zusammenfassung / Abstract

Corynebacterium glutamicum has been used as one of the main producers in fermentation industry, especially for the production of amino acids such as L–lysine. During the past 60 years, molecular biology tools have also been developed for metabolic engineering of C. glutamicum. In this work, a riboswitch–based strategy is used as a promising synthetic biological tool to address this fundamental issue for a more efficient molecular optimization of C. glutamicum for the biosynthesis of L–lysine. The present work has demonstrated a riboswitch–based approach for C. glutamicum metabolic engineering and for the first time investigated a synergetic effect of lysine–ON and lysine–OFF riboswitches for improvement of lysine production in this industrially important microorganism. It is suggested that riboswitches can be effectively used to facilitate strain development in industry.
