This work describes a simple and versatile method for the modification of cell culture dishes with surface-attached, temperature-responsive polymers in which cell adhesion/detachment is controlled by a thermal trigger.
A well-defined series of temperature-responsive polymers that contain photo-crosslinkers is synthesized and characterized. Brief UV irradiation of thin films of the polymers coated onto planar substrates leads to simultaneous crosslinking and attachment of the forming networks to the substrate surfaces. The phase transition behavior of the linear polymers and the corresponding surface-attached hydrogels are studied. Cells cultured onto the polymer modified surfaces show strong changes of cell adhesion as a function of temperature, leading to complete detachment upon briefly cooling to room temperature. A model explaining the protein adsorption behavior and cell adhesion properties of such thermally responsive surfaces is proposed.
