Process Development for the Biotechnological Production and Purification of Mono-, Sesqui- and Diterpenes

Zusammenfassung / Abstract

Terpenes belong to the largest group of natural substances used in the flavoring and food industry up to extremely valuable active ingredients in medicine. In the beginning, they were produced exclusively in plants and trees and had to be extracted using organic solvents. With the far-reaching progress in biotechnology, it is now possible to produce various mono-, sesqui- and diterpenes more sustainably in recombinant microorganisms such as bacteria or yeasts. Here, the production in Escherichia coli was developed with the focus on the integrated isolation and purification of different terpenes. Target molecules were the monoterpenes limonene and perillyl alcohol, the sesquiterpene b-caryophyllene as well as the diterpenes 10-deacetylbaccatin and taxol. These differ extraordinarily in their thermodynamic properties and should therefore represent the product range for process development. The purification method investigated was mainly adsorption chromatography, which, in contrast to extraction, offers advantages in terms of separation performance and sustainability. Furthermore, thermodynamic parameters of the pure terpenes as well as in a variety of liquid phases were simulated based on the COSMO-RS model. Within a short period of time, the simulation enabled an enormous range of reliable values for the thermodynamic characterization of the products as well as the pre-selection of the most suitable solvents in process development.
