Assimilation of one-carbon compounds for the production of bulk chemicals is the primary focus of the third-generation biorefinery concept. Therefore, the anaerobe, Clostridium pasteurianum, was genetically modified and analyzed. The integration of the glycine synthase system from Gottschalkia acidurici led to the consumption of exogenous formate and reduction of native carbon dioxide production. Additional cellular responses were characterized, which were, among others, assessed employing unsupervised learning for phenotype analysis. Furthermore, additional applications were demonstrated for the novel method using fermentation data of C. pasteurianum.
