According to the German Society for Implantology, more than one million implants are placed in Germany every year. Currently, titanium and titanium alloys are predominantly used as implant materials due to their excellent mechanical and biocompatible properties. However, 10% of patients suffer complications, which in the worst case lead to revision surgery. The most common infection that leads to revision surgery is peri-implantitis. Peri-implantitis is a microbial, inflammatory process that occurs in the tissues surrounding dental implants and leads to the loss of supporting bone. In order to curb diseases such as this, the coating of implants with antimicrobial, polymeric biomaterials are being pursued. At the Braunschweig University of Technology, a material has been developed for this purpose that is used as an antimicrobial and self-binding coating for dental implants. This coating was tested in vitro and in vivo: The antimicrobial properties proved to be excellent. The present work has made a significant contribution to the structural elucidation of this polymeric biomaterial. It is produced in a 3-step synthesis and consists, among other things, of an oligoguanidine grafted to polyphosphonates. This goal was pursued on the one hand by substance analysis of all individual synthesis steps and products and on the other hand by surface analysis. Based on the analysis carried out, the following structural proposal was finally formulated for polyPGD (short name of the product). PolyPGD consists of two main components: free PEDBEG and a graft copolymer containing polyD as the main chain. Parts of PEDBEG have been reacted with GMA via ring-opening reactions or aza-Michael reactions. PolyD also contains hemi- and fully hydrolyzed phosphonates. A smaller proportion has been reacted with PEDBEG or PEDBEG-GMA to form phosphonamidates. Non-covalently linked portions in the polymer composition are linked by hydrogen bonds or ionic interactions.
