Negar Rajabi Lab-on-a-Chip for Continuous Cell Perturbation, Lysis and Separation of Subcellular Components
144 Seiten, Dissertation Technische Universität Hamburg-Harburg (2018), Softcover, A5
The dynamic analysis of metabolic processes in compartments of eukaryotic cells is still challenging. This is mainly due to strict requirements of high spatial and temporal analytical resolution, which is difficult to achieve in macro-lab environments. A novel Lab-on-a-Chip is designed, fabricated and characterized for the rapid and automatized preparation of cells for metabolic analysis. The cells are manipulated in the passive microfluidic device solely by the fluid dynamic phenomena caused by the micro channels geometry. In the proposed on-chip cell preparation protocol, cells were perturbed with a metabolic pulse, incubated, and lysed in a continuous flow mode ensuring a high throughput.
