Advancement of instrumentation and novel separation techniques in countercurrent chromatography

Zusammenfassung / Abstract

Countercurrent chromatography (CCC) is a preparative separation technique for the enrichment and the isolation of natural and synthetic compounds. The uniqueness of CCC lies in the usage of two immiscible liquid phases. The advantages of CCC are that irreversible adsorption and denaturation of sample material are mostly avoided. Moreover, the high loading capacity at a low solvent consumption is a further advantage of CCC. On the other hand, this separation technique implies disadvantages with improvement potential. This includes, for example, the low resolving power for structurally similar compounds compared to (preparative) HPLC and the limited choice of suitable solvent systems for the separation of non-polar compounds such as lipids. Novel approaches were developed in this doctoral thesis, the objective of which was to improve the separation efficiency of CCC. On the one hand, the development of novel separation and fractionation techniques was pursued to overcome the difficulties during the separation of lipids by CCC. On the other hand, an improvement of the apparatus was made on a commercially available CCC system by tubing modifications in order to improve the separation efficiency. The developed approaches are also suitable to be used in other CCC instruments.
